Uh well, for starters. Sam Does Not like O'Brien. He thinks he's undisciplined, rude, disrespectful to commanding officers (Carter and his CO) and poor military quality. Sam is used to the rigid, unquestioning obedience that all the Spartans had and being faced with it is something he sees as horribly wrong and downright perverse. He thinks that O'Brien just needs to get slapped around a while and then get dragged into doing things he doesn't want to do so he can shed that entitled attitude.
In the same breath, he's kind of pissed off that O'Brien is so skittish of the Spartans. He hasn't had a whole lot of contact with non-Spartan personnel, so Sam hasn't quite learned of the public opinion yet (especially since they weren't public at all when he kicked it). All the normal soldiers that he's tangled with were pretty fearless, either because they were hand-selected or because they were briefed beforehand on what to expect, whatever. Point is, Sam got something he wasn't expecting and so he made angry faces at it because he's a kid. A competitive, kind of jerky kid sometimes, with an "this is our Spartan Club and if you don't like it you can leave" complex.
He's got absolutely no intention of being nice to O'Brien. ...uh, ever (though Sam's a bit bipolar and stupidly forgiving so if something were to change that he could easily manage an emotional 180). If the guy was about to be killed or was being tortured or something Sam would bust in there, throw O'Brien over his shoulder and get him out, but that's just because that's what he's been trained to do; treat all UNSC soldiers like family. Even if you happen to really dislike that family.

O'Brien would've taken him up on a marksmanship contest, but hand to hand is both his poorest area and a stupidly huge trigger for him.
